go to enterprise manager, expand management then current activity then click on process info...

There is a misunderstanding. I know sp_who and sp_who2. But how I will chek if a specific db is in use except checking users connected or not. Is there any other way?

Maybe you can start a trace using Profiler. Monitor the activity on this one database for as long as is needed to see if anyone's using it. It should be quite easy to set up...look up "profiler" in BOL or ask your questions here and I'm sure you'll get what you need.
